بررسی قابلیت Bifidobacterium bifidumدر جذب آفلاتوکسین M1 از ماست پروبیوتیک

چکیده

توکسین­زدایی میکروبی یکی از روش­های حذف آفلاتوکسین­ها از جمله آفلاتوکسین M1 محسوب می­شود. گزارش­ها نشان دهنده آن است که برخی از سویه­های باکتری­های خانواده اسید لاکتیک از طریق جذب سطحی آفلاتوکسین­ها به دیواره سلولی خود، می­توانند در حذف آن­ها مؤثر باشند. در این تحقیق توانایی باکتری Bifidobacterium bifidum در میزان جذب آفلاتوکسین M1  از ماست پروبیوتیک بررسی گردید. بدین منظور حدود cfu/ml 108 از باکتری پروبیوتیک و آغازگر به شیر پس­چرخ بازسازی شده فاقد آفلاتوکسین M1 تلقیح شد. سپس نمونه­ها با غلظت­های 1/0، 5/0 و 75/0 قسمت در بیلیون (ppb) آفلاتوکسین M1 آلوده شدند. غلظت آفلاتوکسین باقیمانده در سوپرناتانت نمونه­های ماست در روزهای اول، هفتم، چهاردهم و بیست و یکم پس از تولید ماست، توسط روش الایزای رقابتی تعیین و نتایج توسط HPLC تایید شد. یافته­های حاصل ازاین تحقیق نشان داد هرچند، هم باکتری پروبیوتیک مورد آزمون (Bifidobacterium bifidum) و هم آغازگرهای معمولی ماست ( Streptococcus thermophilus و Lactobacillus bulgaricus) قادر به جذب سم می­باشند، توانایی باکتری پروبیوتیک در جذب مقادیر بالای سم در مقایسه با آغازگرهای ماست به لحاظ آماری معنی­دار است. همچنین نتایج حاکی از این است که قابلیت باکتری­ها در کاهش میزان سم در نمونه­های با مقادیر پایین سم بسیار بیشتر است. نتایج حاصل از بررسی زنده­مانی در طی زمان ماندگاری نمونه­های ماست نشان داد که تعداد Bifidobacterium bifidum تا پایان دوره نگهداری ماست (21 روز) در مقایسه با اولین روز حدود 4 سیکل لگاریتمی کاهش یافت.
